Background on ATP Detection

ATP is a ubiquitous organic compound found in all biological materials, including microorganisms and food residues. The measurement of ATP on surfaces can be effectively conducted through luciferase-based luminometer assays, which produce relative light units (RLUs). Low RLU readings indicate effective cleaning, while high readings necessitate further cleaning efforts to ensure hygiene standards are met.


Tianlong’s ATP Hygiene Monitoring System

The Biolum Portable ATP Hygiene Monitoring System produced by Tianlong provides a bioluminescence assay for monitoring hygiene and cleanliness standards when used with ATP Test Swabs (e.g., ATP QuickSwab). Our ATP swabs are pen-sized sampling device that contain recombinant luciferase reagent for detecting the amount of ATP on the basis of admitted fluorescence and to generate visualized results in the form of relative light units (RLUs).

Combined with the testing device, Tianlong Biolum Portable ATP Hygiene Monitoring System is sensitive enough to detect ATP in concentration as low as 10-16 mol within 10 seconds. Moreover, it shows good linearity and sensitivity both in pure analyte studies and ATP from and in presence of food and microbial spoilage.

 

Validation from Independent Laboratory Studies

To objectively validate the performance of the Tianlong system, independent laboratory studies were conducted.

  • Methods of validation

An independent laboratory study was conducted with evaluation of two matrices for stainless steel surface: sweetened yogurt residue and doughnut residue. Prior to application of the matrix, the stainless steel was sanitized by using a UV lamp for 20 minutes, and then cleaned with a 10% Contrad 70 cleaning solution in water followed by a rinse with sterile water. The surface was then washed with 70% isopropanol and allowed to air dry.

For each matrix, 10 replicates were evaluated at the following levels: <10, 10-30, 30-100, 100-300, and 300-1000 RLUs. Each contamination level was homogenized by stomaching for 2 minutes and then spread over a 4”x 4”stainless steel surface area. For the blank level (<10 RLU), an aliquot of sterile water was spread over a 4”x 4”surface area. Regions to be measured allowed to dry for 5 minutes after surface contamination for the wet sampling evaluation, and 1 hour after surface contamination for the dry sampling evaluation. The RLUs were calculated and reported for each level for both sampling times for both matrices.

  • Results Demonstrate Consistency and Accuracy

For the matrix study, all replicates evaluated were within the target RLU levels for both matrices (i.e., sweetened yogurt residue and doughnut residue) and for both wet and dry sampling (Figure 1 & 2).

For each targeted RLU levels, the box and whiskers in the box-plot for each matrix showed compact distribution, indicating highly consistent results across tests, which was greatly benefit from the testing accuracy and stability of our products. It is also worth noting that the detection performance of our products on dry samples was as good as that on wet samples, enabling their flexible application in a variety of production practice scenarios without performance degradation.
